The glucagon-like peptide 1 receptor (GLP1R) is a class B G protein-coupled receptor encoded by the GLP1R gene on human chromosome 6p21.2. It spans the cell membrane seven times and serves as the primary target for glucagon-like peptide-1, an incretin hormone secreted from intestinal L cells in response to nutrient intake. Upon GLP-1 binding, the receptor activates downstream signaling cascades—primarily through Gαs-mediated cAMP accumulation—that promote glucose-dependent insulin secretion, suppress glucagon release, slow gastric emptying, and regulate appetite. The Human GLP1R Stable Cell Line-CHO is created by stably transfecting the full-length human GLP1R coding sequence into Chinese hamster ovary (CHO) cells, a host line chosen for its lack of endogenous GLP-1 receptor expression, robust growth characteristics, and compatibility with high-throughput screening platforms. The resulting clonal population exhibits uniform, stable surface expression of the functional human receptor, enabling consistent and reproducible assay performance across multiple passages.
This cell line is broadly employed in drug discovery programs targeting metabolic diseases. It serves as the primary tool for high-throughput screening of compound libraries to identify novel GLP-1 receptor agonists, including peptide analogs, small molecules, and biased ligands that preferentially activate specific signaling pathways. The line supports a variety of functional readouts, including cAMP accumulation assays, beta-arrestin recruitment measurements, calcium mobilization studies, and receptor internalization analysis. It is also used for binding affinity and kinetic characterization via radioligand or fluorescent ligand competition assays, as well as selectivity profiling against related receptors such as GCGR and GIPR. In addition, the cell line facilitates structure-activity relationship studies during lead optimization, enables safety pharmacology evaluation, and supports mechanistic investigations into GLP-1R desensitization, trafficking, and allosteric modulation.

This CHO GLP1R stable cell line has been an absolute workhorse for our peptide drug discovery assays. The receptor signal-to-noise ratio is excellent, resulting in high Z'-factor values during our primary screening runs. It completely eliminated the batch-to-batch variability we previously faced with transient transfections.
