| Background | Pituitary adenylate cyclase-activating polypeptide type I receptor, also known as PAC1 is a protein that in humans is encoded by the ADCYAP1R1 gene. ADCYAP1R1 is a membrane-associated protein and shares significant homology with members of the glucagon/secretin receptor family. This receptor binds pituitary adenylate cyclase activating peptide (PACAP) mediating several biological activities and it is positively coupled to adenylate cyclase. ADCYAP1R1 is expressed in the adrenal medulla, pancreatic acini, uterus, myenteric plexus and brain. |

The following safety precautions should be observed. 1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum. 2. No eating, drinking or smoking while handling the stable line. 3. Wash hands after handling the stable line and before leaving the lab. 4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells. 5. All waste should be considered hazardous. 6. Dispose of all liquid waste after each experiment and treat with bleach. |

A: The gene editing efficiency in the Human ADCYAP1R1 Stable Cell Line-HEK293 depends on the editing tool used (like CRISPR/Cas9) and the specific target sequence. The efficiency can vary from moderate to very high but needs to be determined experimentally.
A: The expression level of ADCYAP1R1 protein can be quantified through Western Blot or Enzyme-Linked Immunosorbent Assay (ELISA). These methods provide quantitative expression level information.
A: This needs to be determined through drug screening experiments. ADCYAP1R1 is related to multiple signaling pathways, so it might be sensitive to drugs affecting these pathways, such as agonists or antagonists of the G-protein-coupled receptor (GPCR) pathway.
A: Using small interfering RNA (siRNA) or short hairpin RNA (shRNA) are common methods to induce gene silencing. Selecting sequences specific to the ADCYAP1R1 gene can improve silencing efficiency.
A: Mutations in ADCYAP1R1 could affect cell signaling, proliferation, and survival. The specific impacts need to be determined through functional assays like cell proliferation tests, apoptosis analysis, etc.
