The simplest DNA Polymerase known in both size and catalysis.Replication enzyme involved in initiation of DNA replication in eukaryotes.A multi-subunit complex, which inhibits DNA polymerase and primase activities.Isolated from HeLa cells by immunoaffinity chromatography

Used to test in vitro replication capabilities along with other enzymes.Used to test the toxicity of drugs on key human replication systems.Used in DNA replication experiments.

60 mM Tris-HCl (pH 8.0). 5.0 mM magnesium acetate. 0.3 mg/ml bovine serum albumin. 1.0 mM dithiothreitol. 0.1 mM spermine. 0.2 mM of each dCTP, dGTP, dTTP, dATP (pH 7.0). 0.1 mM [α-3H] dATP. 20 μg activated calf thymus DNA. Incubation at 37°C for 30 minutes in a total reaction volume of 30 μl.

One unit is the amount of enzyme required to incorporate 1 nmole of total nucleotide into acid-insoluble form in 60 minutes at 37 °C
