Revival Rapidly thaw cells in a 37°C water bath. Transfer contents into a tube containing pre-warmed media. Centrifuge cells and seed into a 25 cm2 flask containing pre-warmed media.

Safety Considerations The following safety precautions should be observed.

Background This gene encodes the D5 subtype of the dopamine receptor. The D5 subtype is a G-protein coupled receptor which stimulates adenylyl cyclase. This receptor is expressed in neurons in the limbic regions of the brain. It has a 10-fold higher affinity for dopamine than the D1 subtype. Human dopamine receptor D5 (DRD5), historically called D1B, is a member of the rhodopsin-like class A G protein-coupled receptor family. Together with DRD1, it belongs to the D1-like dopamine receptor group, whereas DRD2, DRD3 and DRD4 form the D2-like group. Dopamine is its endogenous agonist. DRD5 principally couples to Gαs, stimulating adenylyl cyclase and increasing intracellular cyclic AMP, which can subsequently activate protein kinase A and other downstream effectors. The human DRD5 gene is located on chromosome 4p16.1, and receptor expression has been reported in neurons within limbic regions of the brain.

The Human DRD5 Stable Cell Line - HEK293 is an engineered model that constitutively expresses human DRD5 in an adherent HEK293 background. It provides a reproducible cellular platform for investigating receptor pharmacology and dopamine-dependent signal transduction without repeated transient transfection. Functional activity can be examined through cAMP accumulation assays, CRE-dependent reporter assays or other validated downstream readouts. The line is suitable for characterizing candidate agonists, partial agonists, antagonists and inverse agonists, generating concentration–response curves and comparing ligand potency or efficacy. It may also support receptor-binding experiments, studies of constitutive activity, desensitization and internalization, and screening programs aimed at compounds that modulate D1-like dopamine signaling.
