| Description | This cell line is a stably transfected cell line which expresses human programmed cell death 1 (PDCD1). |
| Background | PDCD1 protein is a cell surface membrane protein of the immunoglobulin superfamily. It is expressed in pro-B-cells and is thought to play a role in their differentiation. In mice, expression of this gene is induced in the thymus when anti-CD3 antibodies are injected and large numbers of thymocytes undergo apoptosis. Mice deficient for this gene bred on a BALB/c background developed dilated cardiomyopathy and died from congestive heart failure. These studies suggest that this gene product may also be important in T cell function and contribute to the prevention of autoimmune diseases. |

| Safety Considerations |
The following safety precautions should be observed. 1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum. 2. No eating, drinking or smoking while handling the stable line. 3. Wash hands after handling the stable line and before leaving the lab. 4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells. 5. All waste should be considered hazardous. 6. Dispose of all liquid waste after each experiment and treat with bleach. |

A: Gradually adapt to serum-free culture or add specific serum-free growth factors and nutritional supplements to promote cell proliferation.
A: Optimize the choice and dilution ratio of antibodies, extend the incubation time with cells, and ensure that the cells remain viable before analysis.
A: Optimize cell lysis conditions, use efficient immunoprecipitation reagents and appropriate antibodies for PDCD1 protein, ensure sufficient incubation time, and gentle washing steps.
A: Optimize the design of siRNA or shRNA to ensure specificity for the PDCD1 gene, and adjust transfection efficiency and dosage.
A: Ensure uniform experimental conditions, including cell density, drug concentration, and duration of action, and perform multiple repetitions to increase the reliability of the data.
A: Check and optimize culture conditions, reduce physical or chemical stress factors, and use cell protectants to minimize apoptosis.
