Summary Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

scFv(MSLN)-CD28-CD3ζ CAR-T Lentivirus is a gene-delivery reagent developed to generate T cells that recognize mesothelin (MSLN), a cell-surface glycoprotein overexpressed in several solid tumors. The encoded second-generation chimeric antigen receptor contains an extracellular anti-MSLN single-chain variable fragment (scFv), membrane-anchoring elements, the intracellular costimulatory domain of CD28, and the CD3ζ activation domain. The scFv directs antibody-like recognition of surface MSLN without requiring peptide presentation by the major histocompatibility complex. Following antigen engagement, CD3ζ initiates the primary activation signal, while CD28 provides costimulation that supports T-cell activation, proliferation, cytokine production, and effector function. Lentiviral delivery enables efficient gene transfer and stable CAR expression in transduced T cells, creating a practical and reproducible platform for studying MSLN-directed immune responses.

This product is intended for research on mesothelin-targeted cellular immunotherapy and the functional characterization of CD28-based CAR-T cells. It can be used to prepare experimental CAR-T cells for assessing receptor expression, MSLN-dependent activation, target-cell killing, cytokine release, proliferation, and changes in differentiation or exhaustion markers. Comparisons among MSLN-high, MSLN-low, and MSLN-negative target cells can help determine antigen-density requirements, selectivity, and possible on-target effects across different expression levels. The resulting cells are suitable for appropriately designed studies involving malignant mesothelioma, pancreatic cancer, ovarian cancer, esophagogastric cancer, and other MSLN-expressing solid-tumor models. This construct may also serve as a reference for comparing CD28 signaling with first-generation CD3ζ-only receptors or CARs containing alternative costimulatory domains such as 4-1BB. Additional applications include examining soluble or shed mesothelin, repeated antigen exposure, tumor-cell resistance, immune escape, and suppression within the tumor microenvironment. The lentiviral format helps streamline exploratory screening, mechanistic studies, and early-stage evaluation of combination strategies involving checkpoint inhibitors, targeted agents, cytokines, or other immune-modulating approaches.
