Revival Rapidly thaw cells in a 37°C water bath. Transfer contents into a tube containing pre-warmed media. Centrifuge cells and seed into a 25 cm2 flask containing pre-warmed media.

The CEACAM5 gene encodes carcinoembryonic antigen (CEA), a highly glycosylated cell surface glycoprotein belonging to the immunoglobulin superfamily. As a cell adhesion molecule, CEACAM5 mediates interactions between homologous and heterologous cells, thereby influencing cell signaling, differentiation, and survival. CEACAM5 is significantly overexpressed in various human malignancies, including colorectal cancer, gastric cancer, lung cancer, and prostate cancer. High levels of CEA on the cell surface promote tumor cell aggregation and facilitate metastasis by protecting cancer cells from anoikis (cell detachment-induced apoptosis) and enhancing their adhesion to vascular endothelial cells. Furthermore, CEA participates in regulating the immune response in the tumor microenvironment through interactions with receptors on myeloid cells, which often leads to immunosuppression.

A stable human CEACAM5 cell line constructed in the PC-3 context is an ideal model for studying the role of carcinoembryonic antigen in advanced prostate cancer and developing targeted therapies. PC-3 is a human prostate cancer cell line derived from bone metastases, characterized by high invasiveness, androgen-independent phenotype, and high metastatic potential. By stably overexpressing human CEACAM5, this cell line provides a high-density target for evaluating the binding kinetics and efficacy of anti-CEA monoclonal antibodies and therapeutic conjugates.
