The CD276 gene, also known as B7-H3 (B7 Homolog 3), encodes a type I transmembrane protein belonging to the B7 family of immune regulatory molecules. Located on human chromosome 15q24.1, the CD276 protein exists in two isoforms, with the 4Ig-B7-H3 form being predominantly expressed in cancer tissues. CD276 functions as an immune checkpoint molecule, playing a dual role in both adaptive immunity and tumor biology. The Human CD276 Stable Cell Line - CT26 is a stably transfected murine colorectal carcinoma cell line engineered to constitutively express full-length human CD276 on the cell surface. Generated by introducing the human CD276 gene into the parental CT26 cell line via lentiviral or plasmid-based transfection followed by antibiotic selection, this stable cell line provides a reliable and reproducible in vitro model for studying human CD276 in a mouse background. The expression of human CD276 on CT26 cells has been validated by flow cytometry and Western blot, ensuring consistent surface presentation for downstream experimental applications.
This cell line serves as a valuable tool for a wide range of research applications in cancer immunology and drug development. It is particularly useful for evaluating the efficacy and specificity of anti-human CD276 therapeutic antibodies, bispecific antibodies, antibody-drug conjugates (ADCs), and chimeric antigen receptor (CAR)-T cell therapies targeting B7-H3. Researchers can utilize this stable cell line in functional assays such as cell-based binding studies, internalization assays, cytotoxicity tests, and immune cell activation experiments. Additionally, when implanted into immunocompetent syngeneic mouse models, the human CD276-expressing CT26 cells enable in vivo assessment of antitumor immune responses, tumor growth inhibition, and pharmacokinetic/pharmacodynamic profiles of candidate therapeutics targeting CD276. The line also supports mechanism-of-action studies exploring CD276-mediated immune evasion, tumor progression, and resistance to therapy, making it a critical resource for preclinical evaluation of next-generation immunotherapies directed at this promising checkpoint target.

This CD276 stable cell line worked very well in our target-binding and functional assays. Cell recovery was smooth, and the experimental results were reproducible across different assay plates. We found it to be a practical platform for evaluating CD276-targeted candidates.
