| Safety Considerations |
The following safety precautions should be observed. 1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum. 2. No eating, drinking or smoking while handling the stable line. 3. Wash hands after handling the stable line and before leaving the lab. 4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells. 5. All waste should be considered hazardous. 6. Dispose of all liquid waste after each experiment and treat with bleach. |

A: Malignant melanoma cells in humans.
A: The cells have a heavy granular feel. There are more black spots inside the cells and more granules outside the cells during passaging and culture. It is recommended to use high-quality serum and medium to culture the cells.
A: The digestion or resuscitation process should be gentle, strictly control the digestion time and resuscitation operation standards, and it is recommended to observe the cells 24h after passaging or resuscitation, and it is recommended to change the liquid once to remove dead cells.
A: It is recommended to use suitable culture conditions; 2. If there are usually fewer fragments, you can skip the PBS resuspension step when passaging; if there are many fragments, it is recommended to wash with PBS several times.
A: Most of the black particles outside the cells are cell fragments and cell metabolites. You can first suck away the medium and rinse with PBS to remove them, and then add new medium to continue culturing.
A: A375 cells are ideal controls for NRAS mutant-A375 isogenic, suitable for 3D cell culture, high-throughput screening, toxicology, and immunooncology research, and are also suitable transfection hosts.
