| Description | The HIV-1 Activity Assay Kit provides a convenient and simple method for detecting the activity of recombinant and/or purified HIV-1 protease. The assay kit is based on the ability of active HIV-1 protease to cleave a synthetic peptide substrate to release the free fluorophore which can be easily quantified at Ex/Em = 330/450 nm using a fluorescence microplate reader. |
| Component |
•Active HIV-1 protease •HIV-1 protease substrate •HIV-1 assay buffer •FI. standard |
| Applications | Useful for detecting the activity of HIV-1 protease and screening/characterizing the potential inhibitors of HIV-1. |
| Size | 100 assays |
| Method | Fluorescence |
| Caution | Avoid freeze/thaw cycles |
A: HIV-1 protease (PR) is a retroviral aspartyl protease (retropepsin), an enzyme involved with peptide bond hydrolysis in retroviruses, and is critical for the life cycle of HIV.
A: Mature HIV protease exists as a 22 kDa homodimer, with each subunit made up of 99 amino acids.
A: The precursor HIV-1 PR is responsible for catalyzing its own production of the mature PR enzyme through PR auto-processing. The mature protease hydrolyzes peptide bonds at nine specific sites on the Gag-Pol polyprotein, processing the resulting subunits into mature, fully functional proteins.
A: Because HIV-1 protease plays an integral role in HIV replication, it has become a major target for drug therapy.
A: HIV protease inhibitors work by specifically binding to the active site through a tetrahedral intermediate that mimics its substrate and essentially becomes "stuck," thereby inactivating the enzyme. After assembly and budding, virions lacking active proteases are unable to mature into infectious virions.
