Safety and Packaging

Cat. No. CSC-RI0021
Description This cell line is engineered to overexpress human ERG.
Background The FDA, ICH S7B guidelines set out a non-clinical testing strategy to assess the effects of pharmaceuticals on ventricular repolarisation and proarrhythmic risk. The most common mechanism for this is inhibition of the delayed rectified potassium current IKr. The human ether-à-go-go-related gene (hERG) encodes the potassium ion channel responsible for IKr.

Safety Considerations

The following safety precautions should be observed.

1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum.

2. No eating, drinking or smoking while handling the stable line.

3. Wash hands after handling the stable line and before leaving the lab.

4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells.

5. All waste should be considered hazardous.

6. Dispose of all liquid waste after each experiment and treat with bleach.

Q & A

Customer Reviews

Customer Q&As
What is the application of Human ERG Stable Cell Line-CHO in drug cardiotoxicity assessment?

A: This cell line is used to evaluate the effects of drugs on cardiac repolarization, particularly the inhibitory effects on hERG channels (key factors affecting the cardiac delayed rectifier potassium current IKr), to assess the potential risk of arrhythmia.

How to ensure the efficiency of high-throughput screening using Human ERG Stable Cell Line-CHO in drug screening?

A: Utilize automated cell analysis systems and detection methods suitable for high-throughput screening, such as fluorescence-based assays, to improve the efficiency and accuracy of screening.

How to improve the reliability of data in electrophysiological studies using Human ERG Stable Cell Line-CHO?

A: Use precise electrophysiological detection equipment, such as automated patch clamp techniques, ensuring standardized experimental operations and accurate data collection.

How to enhance the detection of specific signals in cell signaling studies using Human ERG Stable Cell Line-CHO?

A: Use antibodies or fluorescent probes specific to the ERG channel, combined with techniques like real-time PCR or Western Blot, to ensure the specificity and sensitivity of the detection signals.

How to reduce false-positive results in drug safety assessment using Human ERG Stable Cell Line-CHO?

A: Perform cross-validation with other cell models or in vitro cardiac models, and use multi-parameter analysis methods to evaluate the comprehensive effects of drugs, thereby reducing false positives.

How to maintain the genetic stability and phenotypic consistency of Human ERG Stable Cell Line-CHO during long-term culture?

A: Regularly perform genetic stability tests, such as karyotyping or gene sequencing, and ensure consistent culture conditions, avoiding over-passaging to maintain phenotypic stability.
