Description SAD-B19 rabies virus-derived retrograde monosynaptic tracer with the native glycoprotein (G) gene deleted and pseudotyped with the avian sarcoma-leukosis virus envelope protein A (EnvA), expressing EGFP. The EnvA pseudotyping restricts initial cellular entry to neurons engineered to express the TVA receptor. Combined with co-injection of an AAV helper expressing the rabies G (RV-G) and TVA in a Cre-dependent or constitutive manner, the virus replicates in starter neurons and crosses a single synapse retrogradely to label directly presynaptic neurons, enabling monosynaptic retrograde input mapping.
