Human neuropeptide Y receptor Y2 (NPY2R or Y2R) is a class A G protein-coupled receptor belonging to the neuropeptide Y receptor family. It is activated by neuropeptide Y (NPY) and peptide YY (PYY), with N-terminally truncated peptides such as PYY(3–36) showing relative preference for Y2 over several other NPY receptor subtypes. NPY2R primarily couples to Gi/o proteins. Receptor activation inhibits adenylyl cyclase and reduces forskolin-stimulated intracellular cyclic AMP, while additional responses may include modulation of ion channels and recruitment of β-arrestins. Y2 receptors are frequently located presynaptically, where they can regulate neurotransmitter release. They are also expressed in central and peripheral tissues and have been studied in connection with appetite regulation, energy balance, gastrointestinal physiology, stress responses and neuronal excitability. These biological roles have made NPY2R an established pharmacological target for both metabolic and neurological research.
The Human NPY2R Stable Cell Line - CHO-K1 is engineered to stably overexpress human NPY2R in Chinese hamster ovary K1 cells. It offers a consistent receptor-expressing background for ligand characterization and GPCR assay development without the experimental variability of repeated transient transfection. Functional activity can be assessed by measuring inhibition of forskolin-induced cAMP accumulation following stimulation with NPY, PYY or receptor-selective peptide fragments. Depending on the assay platform, the cells may also support receptor-binding studies, β-arrestin recruitment measurements, internalization assays or calcium-based detection using an appropriate coupling system. The model is suitable for comparing agonists, partial agonists, antagonists and allosteric modulators, generating concentration–response curves and screening compound libraries for NPY2R activity. It may also assist with studies of receptor desensitization, trafficking and signaling bias.
