The CD19 gene, located on human chromosome 16p11.2, encodes a transmembrane glycoprotein belonging to the immunoglobulin superfamily. Under normal physiological conditions, CD19 expression is restricted to B-lineage cells, from early pre-B cells through mature B lymphocytes, and it functions as a coreceptor that complexes with CD21, CD81, and CD225 to modulate B-cell receptor signaling. The Human CD19/Luc Stable Cell Line - K562 is generated by stably transfecting the K562 erythroleukemia cell line with expression constructs encoding full-length human CD19 and firefly luciferase. The constitutive luciferase expression enables real-time bioluminescent detection, making the cells suitable for both in vitro co-culture assays and in vivo imaging studies.
This cell line serves as a versatile tool for immuno-oncology research, particularly in the development and evaluation of CD19-directed immunotherapies. It is commonly used as target cells in cytotoxicity assays to assess the killing efficacy of CD19-specific CAR-T cells, bispecific T-cell engagers, and antibody-dependent cellular cytotoxicity mediated by anti-CD19 monoclonal antibodies. The bioluminescent signal from the luciferase reporter allows for quantitative, high-throughput readouts in plate-based assays, and facilitates non-invasive tracking of tumor burden in xenograft mouse models. Researchers also employ this line for specificity testing, off-target binding analysis, and functional screening of engineered immune effector cells. By providing a consistent and reproducible CD19-positive target, the cell line supports the preclinical characterization of therapies aimed at B-cell malignancies such as acute lymphoblastic leukemia and non-Hodgkin lymphoma, as well as studies on CD19-directed drug conjugates and combination treatment strategies.

This double-stable K562 line has been a game-changer for our CD19 CAR-T efficacy studies. The CD19 antigen density is uniform, and the luciferase activity shows a linear relationship with cell number, giving us clean, quantitative data in both in vitro killing assays and in vivo imaging.
