Description Cre-conditional, replication-incompetent anterograde monosynaptic HSV-1 H129 tracer. The TK (UL23) gene is replaced by a Cre-activated expression cassette containing a LoxP-Stop-LoxP (LSL) element placed between the strong synthetic CAG promoter and the tdTomato gene. Without Cre, the floxed STOP blocks expression and no tdTomato is produced. In combination with Cre-expressing animals (or local AAV-Cre delivery) and an AAV-DIO-TK helper virus, the STOP is excised specifically in Cre+ starter neurons, allowing monosynaptic anterograde spread to label Cre+ starter neurons and their directly connected postsynaptic neurons.
