Description Cre-conditional, replication-competent anterograde polysynaptic HSV-1 H129 tracer (H356 strain design). The TK locus carries a LoxP-Stop-LoxP-flanked expression cassette linking tdTomato and a codon-modified TK (mTK) via a self-cleaving 2A peptide, all under the strong CAG promoter, with WPRE and poly(A) regulatory elements. Without Cre, the STOP blocks expression. In Cre-expressing neurons, Cre recombinase excises the STOP cassette, restoring TK expression and viral replication; the virus then propagates across multiple synapses anterogradely and labels the entire downstream circuit with tdTomato, providing cell-type-specific polysynaptic tracing.
