Description HSV-1 H129-derived replication-competent anterograde polysynaptic neuronal tracer expressing the red fluorescent protein tdTomato. The tdTomato cassette (under the CMV promoter) is inserted into the H129 genome. Following stereotaxic injection, the virus replicates in infected neurons and propagates anterogradely across multiple synapses, providing bright red fluorescent labeling of multi-order downstream neural networks. The red signal offers excellent spectral separation from EGFP/GFP-labeled counterstains, facilitating dual-color circuit mapping.
