Description HSV-1 H129-derived replication-competent anterograde polysynaptic neuronal tracer expressing enhanced green fluorescent protein (EGFP). The EGFP expression cassette (driven by the CMV promoter) is inserted into the H129 genome. After injection into a target brain region, the virus infects local neurons, replicates, and its progeny virions are transported across multiple synapses in the anterograde direction, enabling visualization of multi-level output neural circuits with bright green fluorescence. HSV-1 H129 has natural anterograde transneuronal spread and strong neuronal tropism, making this an ideal tool for mapping brain-wide output networks.
