What is Caspase-9?

A: Caspase-9 is an enzyme encoded by the CASP9 gene in humans. It is an initiating caspase that is essential for the apoptotic pathway found in many tissues.

What is the structure of Caspase-9?

A: Similar to other caspases, caspase-9 has three domains: an N-terminal pro-domain, a large subunit, and a small subunit.

How does caspase-9 cause apoptosis?

A: When a cell receives signals to undergo apoptosis, caspase-9 is activated through the formation of a protein complex called the apoptosome. Once activated, caspase-9 cleaves and activates downstream effector caspases, such as caspase-3 and caspase-7. These effector caspases then cleave various cellular proteins, leading to the breakdown of the cell's components and ultimately cell death.

How does caspase-9 get activated?

A: In dying cells, activation of caspase-9 is mediated by a multimeric adapter complex called the Apaf-1 apoptosome. The apoptosome is formed by binding of cytochrome c released from mitochondria, ATP, and Apaf-1, which results in the activation of caspase-9.
