Background GLP2 receptor (GLP2R) is a G protein-coupled receptor superfamily member related to glucagon receptor and GLP1 receptor. Glucagon like peptide 2 receptor is expressed in SNC and gut. GLP2R transduces the biological actions of GLP-2, a 33 aminoacid peptide produced by the intestinal endocrine L cell and by various neurons in the central nervous system. Intestinal GLP-2 is co-secreted along with GLP-1 upon nutrient ingestion. GLP-2 produces several actions such as including intestinal growth, enhancement of intestinal function, reduction in bone breakdown and neuroprotection.

Customer Q&As
What is the specific utility of the Human GLP2R-tGFP Stable Cell Line-U2OS in studying G protein-coupled receptors (GPCRs)?

A: The Human GLP2R-tGFP Stable Cell Line-U2OS is a valuable tool for studying the function and regulation of the glucagon-like peptide-2 receptor (GLP2R), a GPCR involved in various physiological processes. By stably expressing the GLP2R fused with a green fluorescent protein (tGFP), researchers can visualize the receptor's subcellular localization and activity in response to ligand stimulation, facilitating studies on receptor trafficking, signaling pathways, and drug interactions.

How does the Human GLP2R-tGFP Stable Cell Line-U2OS contribute to the development of therapeutics targeting GLP2R?

A: This cell line enables high-throughput screening of potential drugs that modulate GLP2R activity. By monitoring changes in tGFP fluorescence, researchers can assess the efficacy of compounds in activating or inhibiting the receptor, aiding in the discovery of new therapeutics for conditions such as diabetes and obesity.

What are the challenges in maintaining the stability of the GLP2R-tGFP fusion protein in the Human GLP2R-tGFP Stable Cell Line-U2OS?

A: Ensuring the long-term expression and functionality of the GLP2R-tGFP fusion protein can be challenging due to factors like gene silencing, chromosomal integration site effects, and proteasomal degradation. Regular monitoring of the cell line's phenotype and functional assays are crucial to maintain its stability.

How does the Human GLP2R-tGFP Stable Cell Line-U2OS aid in understanding the role of GLP2R in cell signaling?

A: By providing a system where GLP2R activity can be directly observed, this cell line allows researchers to investigate the downstream signaling events triggered by receptor activation. This includes studying the involvement of second messenger systems, protein-protein interactions, and gene expression changes.

What are the considerations for using the Human GLP2R-tGFP Stable Cell Line-U2OS in drug safety testing?

A: When using this cell line for safety testing, it's important to consider the potential off-target effects of drugs on the tGFP fluorescence, which could interfere with the readout. Additionally, the cell line's response to drugs should be compared with other cell lines and in vivo models to ensure the relevance of findings.
