| Background | The T-type calcium channel is a type of voltage-dependent calcium channel, the alpha1 subunit determines most of the channel"s properties. The T-type calcium channel is one of the components that produce the pacemaker potential in the SA node of the heart. T-type calcium channel blockers are used primarily as antiepileptics. Two voltage-dependent calcium channels play critical roles in the physiology of cardiac muscle: L-type calcium channel and T-type calcium channels voltage-gated calcium channels. L-type channels are important in sustaining an action potential, while T-type channels are important in initiating them. |

| Revival | Rapidly thaw cells in a 37°C water bath. Transfer contents into a tube containing pre-warmed media. Centrifuge cells and seed into a 25 cm2 flask containing pre-warmed media. |

Wash hands after handling the stable line and before leaving the lab. 4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells. 5. All waste should be considered hazardous. 6. Dispose of all liquid waste after each experiment and treat with bleach. |

A: For cells shipped on dry ice, if the dry ice has completely melted, immediately recover and culture the cells. Do not refreeze. If there is still dry ice, either recover and culture the cells or place the cryovial containing the cells in liquid nitrogen for storage, but do not store the cells in a high-temperature environment.
