Summary Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

The scFv(TACSTD2)-OX40-CD3zeta CAR-T Lentivirus is a recombinant, replication-deficient lentiviral vector designed to redirect T cell specificity toward cells expressing Tumor-Associated Calcium Signal Transducer 2 (TACSTD2), also known as TROP2. The genetic cargo encodes a Chimeric Antigen Receptor (CAR) featuring a specific scFv sequence derived from anti-TROP2 monoclonal antibodies, fused to a transmembrane domain and a potent intracellular signaling complex. This complex integrates the OX40 (CD134) co-stimulatory signaling domain with the CD3-zeta activation domain. By utilizing a lentiviral delivery system, the construct is stably integrated into the T cell genome, ensuring long-term expression and inheritance of the CAR during T cell proliferation. This specific design aims to harness the unique co-stimulatory properties of OX40, which plays a pivotal role in enhancing the survival, expansion, and anti-tumor persistence of engineered T cells when compared to traditional co-stimulatory motifs.

In terms of practical application, this lentivirus is a vital reagent for the development and validation of adoptive cell therapies targeting TROP2-positive solid tumors, including breast, lung, and gastric carcinomas. It is primarily used to transduce primary human T cells or established T cell lines to evaluate the antigen-specific lytic activity and cytokine secretion profiles in co-culture assays with target tumor cells. The product is also instrumental in studying the metabolic reprogramming and phenotypic exhaustion of CAR-T cells, particularly how OX40 signaling modulates the balance between effector and memory subsets. In vivo, these engineered cells are frequently used in systemic or orthotopic tumor models to assess tumor clearance rates and the durability of the immune response. Furthermore, it serves as a reliable benchmark for comparing different CAR architectures or testing synergistic effects when combined with checkpoint inhibitors or other small molecule modulators in the field of cancer immunotherapy research.
