| Summary | Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots. |
| Mycoplasma | Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination. |
| Purity | Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards. |
| Sterility | The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination. |
| Transducibility | Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities. |
| Proviral Identity Confirmation | All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert. |

The scFv(TACSTD2)-CD28-OX40-CD3zeta CAR-T Lentivirus is a specialized genetic engineering tool designed to reprogram human T lymphocytes to target Tumor-Associated Calcium Signal Transducer 2 (TACSTD2), also known as TROP2. This lentiviral vector facilitates the expression of a third-generation chimeric antigen receptor (CAR) on the T cell surface. The construct consists of a high-affinity single-chain variable fragment (scFv) derived from an anti-TROP2 monoclonal antibody, which is linked to a complex intracellular signaling domain. This signaling architecture integrates the primary activation signal from CD3zeta with two potent costimulatory domains, CD28 and OX40. By combining these elements, the CAR is engineered to maximize T cell proliferation, enhance cytokine production, and promote long-term survival. The lentiviral delivery system ensures stable integration of the CAR transgene into the host cell genome, allowing for the generation of a consistent and functional population of therapeutic effector cells.
This lentiviral product is primarily utilized in the development and preclinical evaluation of adoptive cell therapies targeting a wide range of epithelial malignancies. Since TROP2 is highly expressed in various solid tumors, including breast, lung, colonic, and pancreatic carcinomas, this CAR-T construct serves as a pivotal model for studying anti-tumor efficacy in these indications. Researchers employ this lentivirus to perform in vitro cytotoxicity assays, assess cytokine release profiles, and evaluate the metabolic fitness of engineered T cells. Furthermore, the inclusion of the OX40 domain makes it an excellent tool for investigating T cell persistence and the prevention of exhaustion within the challenging immunosuppressive tumor microenvironment. It is also used in the optimization of viral transduction protocols and as a benchmark for comparing the performance of third-generation CAR designs against traditional second-generation constructs in pilot studies and translational oncology research.
