Description Cre-dependent AAV helper expressing a nuclear-localized H2B-BFP reporter together with a codon-modified HSV1 thymidine kinase (cmTK) under EF1a. The optimized cmTK sequence improves expression in mammalian neurons, and the H2B-BFP fusion confines blue fluorescence to the nucleus, giving crisp single-cell resolution of Cre-positive starter neurons during anterograde monosynaptic HSV1-ΔTK tracing.
