Summary Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

EF1a-SV40/LTA(Neo) Lentiviral Particles are high-titer, replication-incompetent viral vectors designed for the robust delivery and stable integration of the SV40 Large T Antigen into mammalian genomes. Controlled by the human Elongation Factor 1 alpha (EF1a) promoter, this system provides consistent and long-term transgene expression across a wide variety of host cells, including difficult-to-transduce primary cells, stem cells, and non-dividing neurons. Unlike common viral promoters such as CMV, the EF1a promoter is highly resistant to transcriptional silencing, ensuring the longevity and stability of the genetic modification over multiple passages. These particles are produced using a secure third-generation packaging system to ensure optimal biosafety and transduction efficiency. Furthermore, the inclusion of a Neomycin (Neo) resistance marker allows for the rapid and efficient selection of stable transformants using G418, making this product an ideal solution for researchers requiring high-fidelity protein expression in complex biological systems.

The primary application of these lentiviral particles lies in the immortalization of primary cell lines, where the SV40 Large T Antigen functions by inactivating the p53 and retinoblastoma (Rb) tumor suppressor pathways. This mechanism allows primary cells to bypass their natural replicative senescence, facilitating the establishment of stable, long-lived cell models that are essential for cancer research, drug discovery, and functional genomics. Additionally, these particles are frequently utilized in the creation of specialized packaging cell lines to enhance the yield of transiently transfected proteins, particularly in systems where the SV40 origin of replication is present. Beyond immortalization, the EF1a-SV40/LTA(Neo) system serves as a critical tool for investigating cell cycle regulation, viral oncogenesis, and intracellular signal transduction. Its capacity to generate permanent, well-characterized cell populations makes it a cornerstone for high-throughput phenotypic screening and the manufacturing of biological therapeutics in both academic and industrial laboratory settings.
