Summary Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

CMV-SV40/LTA(Neo) Lentiviral Particles are high-titer, replication-deficient viral vectors designed for the stable expression of the Simian Virus 40 Large T Antigen (LTA). This system utilizes the robust Cytomegalovirus (CMV) immediate-early promoter to drive high levels of constitutive expression in a wide array of mammalian cell types. The lentiviral delivery mechanism ensures that the SV40 LTA gene is integrated into the host cell genome, providing a permanent genetic modification that remains stable across multiple cell divisions. To assist in the development of uniform cell populations, the particles include a Neomycin resistance marker, which enables the efficient selection of successfully transduced cells through G418 treatment. Unlike standard transfection methods, these lentiviral particles can effectively infect difficult-to-transduce cells, including both dividing and non-dividing primary cultures, making them a versatile tool for long-term genetic studies.

The primary application of these particles is the immortalization of primary mammalian cells. By sequestering and inhibiting the functions of the p53 and retinoblastoma (Rb) tumor suppressor proteins, the SV40 Large T Antigen allows primary cells to bypass replicative senescence and proliferate indefinitely. This capability is crucial for researchers who need to establish stable, continuous cell lines from limited or rare tissue samples while preserving the essential phenotypic traits of the original cells. Beyond immortalization, these particles are frequently used in the production of recombinant proteins and viral vectors, where the presence of the T-antigen can enhance the replication of SV40 origin-containing plasmids. They also serve as a valuable model system for studying oncogenic transformation, cell cycle control, and the molecular pathways involved in DNA replication. The combination of high transduction efficiency and the convenience of Neomycin selection makes these particles an essential resource for creating reliable and reproducible cellular models for biomedical research.
