Calpain Activity Assay Kit

For research use only. Not intended for any clinical use.
Cat.No.
PTAK-036
Description
The Calpain 1 Activity Assay Kit is based on the detection of cleavage of calpain substrate Ac-LLY-AFC. Ac-LLY-AFC possess very weak fluorescence; upon cleavage of the substrate by calpain 1, free AFC can be quantified fluorometrically (Ex/Em = 400/505 nm). In the presence of potent Calpain 1 inhibitor, the hydrolyzation of substrate will be impeded. The kit provides a rapid, simple, sensitive, and reliable test suitable for high throughput screening of Calpain 1 inhibitors. Inhibitor control (Calpain inhibitor 1, ALLN) is included to compare the efficacy of test inhibitors.
Applications
Useful for detecting the activity of calpain 1 and screening the calpain 1 inhibitor.

Avoid freeze/thaw cycles

Customer Q&As
What is calpain?

A: A calpain is a protein belonging to the family of calcium-dependent, non-lysosomal cysteine proteases expressed ubiquitously in mammals and many other organisms.

What is the function of calpain?

A: Calpains are responsible for the controlled proteolysis of specific target proteins, thereby regulating various cellular processes such as cell migration, cytoskeletal remodeling, signal transduction, apoptosis, and synaptic plasticity.

What diseases are calpain associated with?

A: Dysregulation of calpain activity has been implicated in various diseases, including neurodegenerative disorders (e.g., Alzheimer's disease, Parkinson's disease), muscular dystrophy, cardiovascular diseases, and cancer.

What is the size of the calpain?

A: There are two main types of calpain enzymes: calpain-1 and calpain-2. Calpain-1 is a large protein complex consisting of a catalytic subunit (80 kDa) and a regulatory subunit (30 kDa). The full size of the calpain-1 complex is approximately 110 kDa. Calpain-2 is also a large protein complex, similar in size to calpain-1.
