Summary Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

scFv(PSMA)-OX40-CD3ζ CAR-T Lentivirus is a gene-delivery reagent designed to generate T cells that recognize prostate-specific membrane antigen (PSMA), a cell-surface protein frequently expressed at high levels in prostate cancer. The encoded second-generation chimeric antigen receptor combines an extracellular anti-PSMA single-chain variable fragment (scFv) with membrane-anchoring elements, the intracellular costimulatory domain of OX40 (CD134), and the CD3ζ activation domain. The scFv enables antibody-like recognition of surface PSMA without requiring peptide presentation by the major histocompatibility complex. Following antigen engagement, CD3ζ initiates the primary activation signal, while OX40 supplies additional signaling associated with T-cell survival, proliferation, cytokine production, and resistance to activation-induced cell death. Lentiviral delivery supports efficient gene transfer and stable CAR expression in transduced T cells, providing a consistent platform for investigating PSMA-directed immunity and the contribution of OX40 costimulation.

This product is intended for research on PSMA-targeted cellular immunotherapy and the functional evaluation of OX40-based CAR-T cells. Transduced cells can be assessed for receptor expression, PSMA-dependent activation, target-cell killing, cytokine secretion, expansion, survival, and changes in memory or exhaustion phenotypes. Comparisons among PSMA-high, PSMA-low, and PSMA-negative target cells can help define antigen-density requirements, selectivity, and potential on-target effects across different expression levels. The resulting cells are suitable for appropriately designed studies involving androgen-sensitive, castration-resistant, metastatic, and other PSMA-positive prostate cancer models. This construct can also serve as a reference when comparing OX40 with first-generation CD3ζ-only receptors or CARs containing CD28 or 4-1BB costimulatory domains. Additional applications include investigating repeated antigen exposure, antigen loss, tumor-cell resistance, immune escape, and CAR-T-cell performance under immunosuppressive conditions.
