| Summary | Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots. |
| Mycoplasma | Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination. |
| Purity | Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards. |
| Sterility | The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination. |
| Transducibility | Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities. |
| Proviral Identity Confirmation | All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert. |
scFv(Control)-CD28-41BB-CD3ζ CAR-T Lentivirus is a matched control reagent for CAR-T-cell research. It encodes a chimeric antigen receptor containing a control single-chain variable fragment (scFv), membrane-anchoring elements, tandem intracellular costimulatory domains from CD28 and 4-1BB (CD137), and the CD3ζ activation domain. The control scFv is selected to lack relevant recognition of the target antigen used in the experimental system, while retaining an overall receptor architecture comparable to that of an antigen-specific third-generation CAR. This design helps separate effects caused by target recognition from those related to lentiviral transduction, CAR expression, or intracellular signaling components. Lentiviral delivery supports efficient gene transfer and stable receptor expression in transduced T cells, providing a consistent control population for side-by-side experimental comparisons.
This product is intended for use as a negative control in studies of antigen-specific CAR-T-cell activity. T cells transduced with the control lentivirus can be compared with target-specific CAR-T cells when measuring transduction efficiency, receptor expression, cell viability, proliferation, differentiation, basal activation, and expansion. In co-culture assays, the control cells help establish background levels of target-cell killing, cytokine secretion, degranulation, and nonspecific responses to antigen-positive or antigen-negative cells. The construct is also useful for evaluating tonic signaling, activation caused by receptor clustering, and cellular changes associated with prolonged CAR expression. In animal studies, an appropriately validated control group may help distinguish antigen-dependent effects from responses caused by cell administration, vector exposure, culture conditions, or the tumor model itself. Because the intracellular architecture includes both CD28 and 4-1BB, the reagent is especially suitable as a backbone-matched control for third-generation CAR constructs containing the same signaling domains. Its lentiviral format supports reproducible control-cell preparation across screening, mechanistic, and preclinical research workflows.
