| Safety Considerations |
The following safety precautions should be observed. 1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum. 2. No eating, drinking or smoking while handling the stable line. 3. Wash hands after handling the stable line and before leaving the lab. 4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells. 5. All waste should be considered hazardous. 6. Dispose of all liquid waste after each experiment and treat with bleach. |

A: It was created by genetically modifying the mouse MC38 cell line using a lentiviral vector carrying the human FGL1 sequence, resulting in a stable MC38 clone expressing exogenous human FGL1.
A: Primarily used for drug screening and biological assays, especially in studying the interaction of FGL1 with the immune checkpoint molecule LAG3 and its effect on antigen-specific T cell activation.
A: It requires specific culture conditions, including RPMI1640 medium with puromycin and appropriate cell splitting ratios.
A: Its importance lies in studying the role of FGL1 in tumor immune evasion and immune checkpoint therapies due to FGL1's involvement in immune suppression.
A: Useful for assessing drugs targeting the FGL1 and LAG3 interaction and their impact on antigen-specific T cell activation.
A: Cells should be stored in liquid nitrogen and properly resuscitated and cultured prior to use, maintained under recommended conditions.
