Description Cre-conditional, replication-incompetent anterograde monosynaptic HSV-1 H129 tracer (H361 strain design). The TK gene is deleted and replaced by a Cre-activated LoxP-Stop-LoxP (LSL) cassette driving tdTomato expression. Without Cre, no tdTomato is produced and the virus cannot replicate. When combined with Cre-expressing animals and an AAV-DIO-TK helper virus, Cre excises the STOP, tdTomato is expressed in Cre+ starter neurons, and TK complementation from the helper AAV enables a single round of replication and anterograde spread across one synapse, specifically labeling the direct postsynaptic targets of Cre+ neurons.
