Description Replication-incompetent anterograde monosynaptic HSV-1 H129 tracer with the thymidine kinase (TK, UL23) gene deleted. The tdTomato reporter is expressed under the human ubiquitin C (hUbC) promoter. Because TK is essential for viral genome replication in neurons, this tracer cannot replicate or cross synapses on its own. When co-injected at the same site with an AAV helper virus (e.g., AAV-EF1α-TK) that provides TK in trans, the tracer replicates only in AAV-TK-transduced starter neurons and crosses a single synapse to label directly postsynaptic neurons, enabling precise monosynaptic output mapping.
