The following safety precautions should be observed. 1. Use pipette aids to prevent ingestion and keep aerosols down to a minimum. 2. No eating, drinking or smoking while handling the stable line. 3. Wash hands after handling the stable line and before leaving the lab. 4. Decontaminate work surface with disinfectant or 70% ethanol before and after working with stable cells. 5. All waste should be considered hazardous. 6. Dispose of all liquid waste after each experiment and treat with bleach. |

Gamma-aminobutyric acid (GABA)-gated ion channels are widely distributed in the mammalian brain and are major mediators of inhibitory synaptic transmission. A typical GABA ion channel has a pentameric structure consisting of 5 protein subunits, often alpha, beta and gamma or delta, combining to form a central ion conducting pore across the cell membrane. In humans there are six genes that encode alpha subunits, three that encode beta, three that encode gamma, and an additional seven genes that encode other subunits whose function is less-well understood than the alpha, beta and gamma subunits. GABA ion channels open and close in response to secretion of GABA from presynaptic terminals. GABAA alpha4 channels comprised of alpha4, beta3, and gamma2 subunits are extrasynaptic, estimated to represent<5% of="" all="" gabaa="" receptors="" located="" in="" the="" brain,="" and="" insensitive="" to="" benzodiazepines.="" research="" interest="" has="" focused="" on="" alpha4-containing="" receptors="" because="" they="" are="" potentiated="" by="" concentrations="" of="" ethanol="" experienced="" by="" human="" drinkers="" (1-3="" mm);="" one="" study="" showed="" this="" effect="" requires="" the="" delta="" subunit="" is="" part="" of="" the="">5%>
A: This cell line is based on HEK293 cells, with the human GABRA4 (gamma-aminobutyric acid A receptor alpha4 subunit) gene stably transfected into these cells.
A: It is mainly used for studying the role of GABRA4 in neural system functions, especially in neural transmission and emotional regulation.
A: It can be used for screening drugs that affect GABRA4 activity, particularly in the treatment of neurological diseases and mood disorders.
A: The cells should be handled under sterile conditions, with regular monitoring of cell growth and GABRA4 protein expression levels.
A: They should be cultured at 37°C in a 5% CO2 environment, using a medium suitable for the growth of HEK293 cells.
A: It can be used to study the effects of various drugs on the GABRA4 receptor, thereby understanding the mechanisms of drugs in treating neurological diseases.
