| Summary | Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots. |
| Mycoplasma | Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination. |
| Purity | Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards. |
| Sterility | The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination. |
| Transducibility | Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities. |
| Proviral Identity Confirmation | All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert. |
VDR/DR3 Luciferase Reporter Lentivirus is a pathway-responsive gene-delivery reagent designed to measure transcriptional activation mediated by the vitamin D receptor (VDR). The lentiviral construct contains a luciferase reporter gene controlled by a minimal promoter and vitamin D response elements arranged in the DR3 configuration, in which two nuclear-receptor recognition motifs are separated by three nucleotides. When an active vitamin D ligand, such as 1,25-dihydroxyvitamin D3, binds VDR, the receptor undergoes a conformational change and forms a heterodimer with retinoid X receptor. The activated VDR–RXR complex binds DR3-type response elements, recruits transcriptional coregulators, and promotes luciferase expression. The resulting luminescent signal provides a sensitive and quantitative measure of VDR-dependent transcription. Lentiviral delivery supports efficient transduction and stable reporter integration, making the system suitable for transient pathway studies as well as the generation of VDR-responsive stable cell lines.
This reporter lentivirus can be used to investigate vitamin D signaling, nuclear-receptor regulation, and the transcriptional effects of natural metabolites or synthetic compounds. In transduced cells, changes in luminescence allow researchers to generate concentration-response curves, compare ligand potency and efficacy, and characterize the kinetics of VDR activation. The system is well suited for screening VDR agonists, partial agonists, antagonists, and vitamin D analogs, as well as compounds that influence receptor expression, ligand metabolism, heterodimer formation, or coregulator recruitment. It can also support studies of calcium and phosphate homeostasis, bone metabolism, epithelial differentiation, immune regulation, and other biological processes influenced by VDR signaling. Additional applications include evaluating genetic perturbations of VDR pathway components, investigating receptor variants, studying pathway cross-talk with other nuclear receptors, and comparing responses among different cellular backgrounds.

The luciferase signal was exceptionally strong and consistent in our VDR ligand screening assays. Even at low MOI, we detected clear dose-dependent responses.
