Summary Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

The TR/DR4 luciferase reporter lentivirus is a third-generation self-inactivating lentiviral system engineered to deliver a firefly luciferase gene driven by a minimal promoter. This promoter is controlled by multimerized thyroid hormone receptor response elements arranged as direct repeats (DR4) separated by four nucleotides. This structure confers high specificity to TRα/β (which typically forms heterodimers with RXR) while maintaining very low background activity, thereby maximizing the signal-to-noise ratio and dynamic range. VSV-G pseudotyping supports a broad host range and enables efficient transduction of both dividing and non-dividing cells, achieving stable genomic integration and long-term, copy number-dependent reporter gene expression in various cell types and culture models. To enhance transcriptional stability and improve expression robustness without increasing background signal, the vector typically includes post-transcriptional regulatory elements (e.g., WPRE) and a polyadenylation signal downstream of the luciferase ORF. The self-inactivating 3'' LTR, removal of accessory genes, and production using a separate packaging system ensure that the viral particles are replication-incompetent, prioritizing safety and reliability.

This reporter system is widely applicable for quantitative analysis of thyroid hormone signaling in basic research and translational medicine workflows. In pharmacology and drug discovery, it supports high-throughput screening of TR agonists, partial agonists, and antagonists, enabling potency ranking and EC50/IC50 determination with rapid turnaround times and clear background separation. Because the DR4 motif binds to the TR/RXR heterodimer, the system is well-suited for studying pathway crosstalk and combination pharmacology with retinoids, co-regulator modulators, or epigenetic drugs. It facilitates structure-activity relationship studies and mechanism of action analysis, including assessment of ligand preference, chromatin environment dependence, and co-activator/co-repressor usage. In the field of cell biology, lentiviral vector systems allow for the creation of stable reporter gene cell lines in disease-relevant models (such as hepatocytes, cardiomyocytes, neurons, adipocytes, thyroid-derived cell lines, iPSCs, and organoids) to monitor endogenous pathway status, induction kinetics after T3/T4 administration, and adaptation to hormonal environments using hormone-deficient culture media.

Exceptional Signal Sensitivity

The luciferase signal from this lentivirus was remarkably strong and specific in our TR/DR4 pathway studies. Even at low cell densities, we detected clear responses.
