Summary Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

The SP1 luciferase reporter lentivirus is a versatile, next-generation transcriptional reporter system designed for high sensitivity, stability, and broad cell compatibility. It contains a luciferase reporter gene driven by a minimal promoter located downstream of a multimerized SP1 response element, enabling low basal activity and high inducibility upon SP1 (Specificity Protein 1) activation. The lentiviral vector ensures stable genomic integration in both dividing and non-dividing cells, guaranteeing long-term stable expression and consistent detection results in passaging and time-course studies. The luciferase-generated luminescence signal offers high sensitivity, allowing for linear quantification across several orders of magnitude and compatibility with standard luminometers for rapid, scalable measurements.

This reporter system is ideally suited for applications requiring real-time or long-term quantitative analysis of SP1 pathway dynamics. It can be used to analyze pathway activation in different cell types, assess lot-to-lot consistency of upstream reagents, or perform dose-response characterization of small molecules, biologics, and genetic perturbations that modulate SP1 activity. Because SP1 regulates many genes with GC-rich promoter elements, this reporter system is valuable in oncology for studying proliferative signaling, in toxicology and oxidative stress research for studying SP1''s response to cellular stress, and in inflammation and metabolic research for studying SP1-driven pathways in transcriptional reprogramming. The virus seamlessly integrates into high-throughput screening workflows for drug discovery and lead compound optimization, enabling rapid screening of hit compounds based on potency, efficacy, and kinetics.
