Summary Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

The scFv(Control)-CD28-CD3zeta CAR-T Lentivirus is a standardized tool designed for the rigorous validation of second-generation CAR-T cell therapies. This lentiviral vector facilitates the expression of a chimeric antigen receptor that incorporates a non-targeting single-chain variable fragment (scFv) as its extracellular sensing element. This mock scFv is fused via a hinge and transmembrane region to the CD28 co-stimulatory endodomain and the CD3ζ primary signaling domain. By utilizing a high-titer lentiviral delivery system, the construct is stably integrated into the host T-cell genome, ensuring persistent expression across multiple cell divisions. This specific configuration allows researchers to maintain the structural and intracellular signaling integrity of a standard second-generation CAR while eliminating antigen-specific recognition, providing a crucial benchmark for therapeutic assays.

The applications of this control lentivirus are central to preclinical development and mechanistic studies. It is primarily employed as a negative control in cytotoxicity assays, such as chromium release or luciferase-based killing assays, to accurately subtract background cell lysis from targeted, antigen-driven lytic activity. Beyond simple killing assays, it serves as a vital reference for assessing “tonic signaling,” a phenomenon where CAR-T cells exhibit spontaneous activation and exhaustion in the absence of an antigen. By comparing the baseline cytokine production and phenotypic markers of cells transduced with this control virus against those of antigen-specific CAR-T cells, scientists can isolate the metabolic and transcriptional changes triggered solely by antigen binding. Additionally, it is used to validate that any observed off-target effects or systemic toxicities in in vitro models are not inherent to the viral transduction process or the constitutive expression of the CD28-CD3ζ signaling scaffold.
