Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

The RAR/DR5 luciferase reporter lentivirus is a versatile, ready-to-use reporter system designed to transduce a variety of mammalian cells and provide stable, sensitive readout of the retinoic acid receptor (RAR) signaling pathway. Its core design places the firefly luciferase gene under the control of a minimal promoter driven by tandem RAR response elements (DR5), ensuring extremely low background activity and a high dynamic range upon ligand-dependent activation. Because the lentiviral vector backbone integrates into the host genome, the reporter gene signal remains stable throughout long-term culture and cell division, allowing for the creation of stable reporter cell lines without repeated transfection. This stability is particularly advantageous for difficult-to-transfect cell types, including primary cells and slowly dividing cell populations, ensuring consistent data across diverse biological models.

This reporter system is broadly applicable to various applications requiring quantitative assessment of retinoid pathway activity. It allows for direct measurement of the effects of all-trans retinoic acid, synthetic retinoids, or pathway modulators on RAR-driven transcription, supporting potency ranking, structure-activity relationship exploration, and selectivity analysis against other nuclear receptor families. In basic research, it is ideal for dissecting RAR signaling dynamics in developmental biology, differentiation trajectories (e.g., neuronal or epithelial lineages), and tissue homeostasis, as well as studying interactions with RXR, Wnt, TGF-β, MAPK, and other pathways. In drug discovery, the assay can be used for primary and secondary screening, counter-screening for off-target activation, and mechanism of action studies, and can be easily integrated into automated luminescence detection systems for high-throughput, dose-response, and time-course analysis.
