The pCDisplay-4 vector is a highly versatile and widely used tool in the field of molecular biology research and genetic engineering. The promoter sequences that drive gene expression in the pCDisplay-4 vector are M13 and ColE. Promoters are essential elements in the process of transcription, these sequences derived from bacteriophages and colicins are known for their robust and efficient gene expression, enabling high yield of the proteins of interest. One of the essential features of the pCDisplay-4 vector is its resistance to ampicillin, which is the primary selection criterion. Following transformation, the host cells are cultivated in a medium containing ampicillin. Those harboring the pCDisplay-4 vector would have acquired ampicillin resistance and will survive, while the others will die. This forms the basis of a powerful selection methodology ensuring that only cells containing the vector grow and divide. The pCDisplay-4 vector is labeled with two tags, His6 and HA tag. The presence of these tags allows easy identification and purification of the proteins of interest. His6 tag is a string of six histidine residues used to bind proteins to certain metal ions commonly used in protein purification columns. On the other hand, HA tag is a human influenza hemagglutinin tag, it’s widely used due to its small size and can be recognized by commercially available antibodies for both detection and purification of tagged proteins.

CD133, also known as Prominin-1, is expressed on stem cells in many tissues and tumors. In this work, researchers identified and characterized a single-chain variable fragment (scFv) for efficient and specific recognition of CD133.

Here, researchers used the VL and VH fragments of clone 7 anti-CD133 antibody to construct recombinant anti-CD133 scFv. VL and VH fragments were amplified by RT-PCR. Using these fragments and a DNA linker encoding Ser7Gly10Arg, scFv was constructed by performing overlap extension PCR. The resulting PCR products were analyzed by gel electrophoresis (Figure 1a). The scFvs were then cloned into the pCDisplay-4 vector, transformed into E. coli XL-1 BLUE, and subsequently infected with VCSM13 helper phage to release intact phage particles displaying the scFv library on their coat surface. The phage library was then screened for reactivity to CD133 by a standard panning process that involves incubating phage particles with CD133 protein-coated plates. Only a small fraction of the phage library was found to bind to the plate. These conjugates are then recovered, amplified in bacteria and screened by subsequent panning. After 3 such cycles, the phage conjugates were enriched 394-fold (Figure 1b). At this stage, 30 phage clones were randomly picked and their reactivity toward human CD133 was confirmed by phage ELISA (Figure 1c). The results show that CD133 scFv can recognize and bind to human CD133 protein, but the binding affinity is lower than its parent monoclonal antibody.

The pCDisplay-4 vector, a type of phagemid vector, has various applications in the field of molecular biology, genetics and biotechnology. Primarily, pCDisplay-4 vectors are extensively used in the construction of expression libraries, particularly in the construction of phage display libraries. Herein, their application enables the display of large peptide and protein libraries on the surface of filamentous phages. These libraries can then be used to map epitopes and identify interaction sites on proteins, as well as to screen for novel peptides or proteins that bind a particular target molecule. In addition, these libraries can also be utilized to study protein-protein, protein-DNA, and protein-RNA interactions. pCDisplay-4 vector use in molecular biology offers significant advantages which include its stability, ease of propagation, high transformation efficiency, and restriction sites that allow for straightforward subcloning. Furthermore, the fusion proteins displayed on the phage surface are physically linked to the gene that encodes them, allowing for the rapid identification of interacting proteins. More recently, the pCDisplay-4 vector has been playing a pivotal role in the field of therapeutic protein discovery and development. Multiple successful therapeutic antibodies have been discovered using phagemid-based display technologies, which prove the importance of pCDisplay-4 vector in modern biotechnology.
