Summary Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

Customer Reviews

The MEF2 luciferase reporter lentivirus is a powerful research tool composed of lentiviral particles carrying a luciferase reporter gene driven by a minimal promoter and a MEF2 response element. This viral vector system offers several key advantages for studying the MEF2 (myocyte enhancer factor 2) signaling pathway. The lentiviral vector stably integrates into the host genome, allowing for long-term expression of the reporter construct in both dividing and non-dividing cells. The system exhibits high transduction efficiency in a wide range of cell types, including primary cells and neuronal cells that are difficult to transfect. The minimal promoter ensures low basal activity while maintaining high sensitivity to MEF2 transcriptional regulation, allowing researchers to specifically monitor MEF2 activity without interference from other signaling pathways.

The MEF2 luciferase reporter lentivirus has broad applications in biomedical research, particularly in studies of muscle development, neuronal differentiation, and cardiac biology, where the MEF2 transcription factor plays a crucial role. Researchers utilize this system to investigate how various stimuli, such as growth factors, stress signals, or drug compounds, regulate MEF2 transcriptional activity. In drug discovery, it serves as a screening platform to identify compounds that activate or inhibit MEF2-dependent pathways. This lentiviral reporter is particularly valuable for studying the dynamic changes of MEF2 during cell differentiation in stem cell research. Neuroscientists use this tool to investigate the role of MEF2 in synaptic plasticity, neuronal survival, and activity-dependent gene regulation. Furthermore, the system can be used to explore the mechanisms of diseases involving MEF2 dysregulation, including muscular dystrophy, neurodegenerative diseases, and cardiovascular diseases.
