NECTIN4, also known as Nectin cell adhesion molecule 4, is the protein encoded by the NECTIN4 gene in humans. This protein belongs to a family of cell adhesion molecules (CAMs) called nectins, which are involved in a range of biological processes such as cell migration, proliferation and differentiation. Nectin is best known for its role in the formation of adherens junctions, which are essential for maintaining tissue structure and function. Specifically, NECTIN4 has been implicated in the development and function of the nervous and immune systems. Recent studies have also suggested that NECTIN4 may function as a tumor marker, as it is frequently overexpressed in various types of cancer, including breast, ovarian, and lung cancers. Therefore, NECTIN4 is considered a potential target for cancer treatment.
The CHO-K1 (Chinese Hamster Ovary-K1) host cell line is derived from the ovaries of Chinese hamsters and is specifically designed for protein production. Due to their efficient gene expression mechanism, they are widely used in biological and medical research. Human NECTIN4 Stable Cell Line - CHO-K1 is a cell line genetically engineered to stably express the human NECTIN4 gene. These cell lines are important tools for biomedical research, particularly in studying the role of the NECTIN4 gene in cancer initiation and progression and potential drug development.
Nectin-4, also known as PVRL4 (poliovirus receptor-like 4), is specifically expressed in the embryo and placenta. Recent studies have reported that Nectin-4 is overexpressed in a variety of human cancers, and this aberrant expression is associated with cancer progression and poor patient prognosis. Here, researchers first examined the expression of Nectin-4 in human esophageal cancer (EC) tissues using immunohistochemistry (IHC) assays and analyzed the clinical relevance. In vitro cell studies and nude mouse in vivo tumor models were then used to examine the regulatory role of Nectin-4 in EC progression. The results showed that overexpression of Nectin-4 in human EC tissues was significantly associated with tumor size, tumor invasion depth, and poor patient prognosis. Intervention of Nectin-4 expression in EC cell lines showed that increased Nectin-4 expression could significantly promote cell viability, migration, invasion, and tumor formation. Therefore, Nectin-4 plays a crucial role in tumor biology and it can serve as a useful prognostic predictor for human EC.
Figure 1. Intervention of Nectin-4 expression affects the migration ability of human EC cell lines. The effect of intervention of Nectin-4 expression on the migration ability of human EC cell lines was evaluated by scratch healing assay. a The migration ability of Eca-109 and TE-1 cells was significantly reduced after knockdown Nectin-4 expression, and the cell-free area of the LV-Nectin-4-shRNA group was significantly larger than that of the LV-NC group at 24 h. b The migration ability of Eca-109 and TE-1 cells was significantly enhanced after overexpression of Nectin-4, and the cell-free area of the LV-Nectin-4-OE group was significantly smaller than that of the LV-Vector-Ctrl group at 24 h. (Deng H, et al. 2019)
Creative Biogene has successfully constructed NECTIN4 over-expressing stable cell lines with CHO-K1, HEK293, Eca-109, TE-1, CHO, HeLa, 1321N1, U2OS, RH7777, etc. These NECTIN4 stable cell lines have excellent in vitro assay sensitivity and reproducibility. Therefore, conducting experiments with our NECTIN4 stable cell lines saves time and resources while providing more accurate results.
