CD27, a key member of the tumor necrosis factor receptor (TNFR) superfamily, plays a critical role in regulating immune responses. CD27 is primarily expressed on T cells, B cells, and natural killer (NK) cells. As a co-stimulatory molecule, it enhances lymphocyte activation, proliferation, and survival. Its interaction with its ligand CD70 provides key signals for the generation and maintenance of T cell memory and the differentiation of plasma cells. Dysregulation of the CD27-CD70 axis is associated with a variety of immune-related diseases, including autoimmune diseases and cancer, making it a promising therapeutic target.
Human CD27 virus-like particles (VLPs) are innovative biomimetic nanoparticles designed to mimic the structural and functional properties of native viruses while removing viral genetic material to ensure safety and biocompatibility. These VLPs are specifically designed to display CD27 molecules on their surface, thereby exerting the immunostimulatory effects of CD27 in a multivalent format. The repetitive arrangement of CD27 on the VLP enhances its ability to bind to CD70-expressing immune cells, thereby amplifying the co-stimulatory signal and promoting robust immune activation. CD27 VLPs have great potential as vaccine adjuvants or stand-alone immunotherapies, particularly for cancer and infectious diseases, where eliciting durable T cell responses is crucial.
