CD22, also known as cluster of differentiation 22 or SIGLEC-2, is a transmembrane glycoprotein belonging to the sialic acid-binding immunoglobulin-like lectin family. It is predominantly expressed on the surface of mature B cells and plays a key regulatory role in B cell receptor signaling by modulating immune activation thresholds. The Human CD22/Luc Stable Cell Line - K562 is an engineered cell line derived from the K562 parental line, which originates from a human chronic myelogenous leukemia patient. These cells have been stably transduced to co-express full-length human CD22 on the cell surface along with a firefly luciferase reporter gene. The luciferase activity enables real-time bioluminescent detection, making these cells suitable for both in vitro and in vivo imaging applications.

This cell line serves as a versatile tool in immuno-oncology research, particularly for evaluating therapeutics targeting CD22. It is widely used in CAR-T cell cytotoxicity assays, where CD22-directed CAR-T cells are co-cultured with the target cells and killing efficiency is measured via luciferase-based readouts. The bioluminescent signal directly correlates with viable cell numbers, allowing for quantitative, high-throughput assessment of target cell lysis. Beyond CAR-T development, the line supports antibody-dependent cellular cytotoxicity (ADCC) assays, bispecific T cell engager (BiTE) evaluations, and antibody-drug conjugate (ADC) screening. Researchers also employ it for in vivo tumor xenograft models, where luciferase imaging permits non-invasive tracking of tumor burden and therapeutic response over time. Additionally, the cell line is useful for studying CD22 internalization dynamics, receptor-mediated signaling, and B cell biology in a controlled, non-B-cell context. Its stability, ease of culture, and compatibility with standard bioluminescence platforms make it a practical choice for both academic and industrial drug discovery programs.

A Reliable Tool for CD22 CAR-T Cytotoxicity Assays

Our lab uses this line routinely for CD22 CAR-T co-culture experiments. CD22 surface expression is consistent across passages, and the luciferase signal remains bright and stable, making the readout highly reproducible. Thawed cells recovered quickly with excellent viability, and the lot-to-lot consistency has saved us countless optimization hours.
