Stable cell line of monkey CD47 With regard to regulating intracellular calcium concentration following cell attachment to the extracellular matrix, CHO-K1 is a cell line that has been modified to express CD47. The C-terminal cell binding domain of thrombospondin is likewise a receptor for CD47, which may have an effect on signal transduction and membrane transport. This gene is widely distributed throughout tissues and is expressed less on Rh erythrocytes. For CD47, many variations of alternatively spliced transcripts have been found. CHO-K1 is a cell line derived from Chinese hamster ovary cells widely used in biotechnology and biomedical research. These cells can be used to express recombinant proteins because of their inherent growth. When cultivated under the right conditions in conventional cell culture medium, CHO-K1 cells offer a flexible platform for researching gene expression and protein function.

Treatment of Diffuse Large B Cell Lymphoma (DLBCL) patients using rituximab and the CHOP regimen commonly encounters challenges due to both inherent and acquired resistance mechanisms. The researchers investigated the novel bispecific fusion protein DSP107 for treating DLBCL. DSP107 targets the CD47:SIRPα signaling axis to enhance phagocytosis by macrophages and trogocytosis by PMNs against DLBCL cells. It also activates 4-1BB-mediated costimulatory signaling in T cells, boosting their anticancer activity. In vitro and in vivo studies demonstrated DSP107's ability to significantly reduce cancer cell numbers and tumor size, particularly when combined with rituximab. Safety evaluations in cynomolgus monkeys showed a favorable profile. These findings suggest that DSP107 is a promising therapeutic strategy for enhancing innate and adaptive immune responses in DLBCL treatment.
