A: One unit is defined as the amount of enzyme required to completely digest 1 μg of λ DNA in a total reaction volume of 50 μl within 1 hour at 65°C.
A: BsiHKC I recognizes the C↓PyCGPuG site and works best at 65°C but does not function at 37°C.
A: BsiHKC I is supplied with CMX Buffer 3, which provides the highest activity. CMX Buffer 2 or CMX Buffer 4 can be used as alternatives if CMX Buffer 3 is not available, but the effect is not as good as with Buffer 3.
A: CMX Buffer 3 for BsiHKC I contains 10 mM Tris-HCl (pH 7.5 at 37°C), 50 mM NaCl, 10 mM MgCl, 21 mM DTT, and 100 μg/ml bovine serum albumin (BSA).
