Species SV40
Application SV40 small/large T antigen lentivirus (EF1α, Puro) is a versatile tool used primarily in molecular and cell biology research. Here are some of its key applications: Cell line immortalization: SV40 large T antigen is able to inactivate tumor suppressor proteins such as p53 and retinoblastoma protein (pRb), allowing cells to bypass normal cellular senescence and continue to divide, effectively immortalizing primary cells. This is very useful for creating stable, long-term cell cultures for research. Gene expression studies: The use of the EF1α promoter ensures that SV40 small/large T antigen is stably and consistently expressed in a variety of cell types, facilitating gene regulation and expression studies under controlled conditions. Cancer research: Since SV40 large T antigen is known to interfere with tumor suppressor pathways, it can be used to create models to study cancer biology, tumorigenesis, and the underlying mechanisms of cell cycle regulation and apoptosis. Drug screening and development: Immortalized cell lines generated using SV40 T antigen can be used in high-throughput screening assays to evaluate the efficacy and toxicity of potential therapeutic compounds. Pluripotent stem cell research: SV40 T antigen helps maintain the undifferentiated state of pluripotent stem cells, which is critical for stem cell research and regenerative medicine.

Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

Simian virus 40 (SV40) is a well-known polyomavirus that has made a great contribution to the study of virology and molecular biology. Among the proteins encoded by SV40, the small T antigen (ST) and large T antigen (LT) play crucial roles in viral replication and cellular transformation. These proteins interact with a variety of cellular targets and affect cell proliferation, transformation, and apoptosis. Manipulation of the SV40 T antigen using a lentiviral system allows for powerful experimental approaches to study these fundamental processes in eukaryotic cells. Lentiviral vectors are widely used for gene delivery due to their ability to stably integrate into the genome of both dividing and non-dividing cells. Integration results in long-term expression of the delivered gene. Expression of SV40 T antigen using lentiviral vectors provides a powerful and efficient method to study its role in various cell types. The SV40 small/large T antigen lentivirus (EF1α, Puro) is a powerful genetic tool designed to express untagged SV40 small T and large T antigens in mammalian cells. Lentivirus utilizes the EF1α (elongation factor-1 alpha) promoter, which ensures stable and consistent expression of the antigen in a variety of cell types, including dividing and non-dividing cells.
Customer Q&As
What starting range of puromycin should I use for the antibiotic selection?

A: The concentration must be optimized case-by-case and is cell type dependent. On average, typical selection amounts are around 0.1 to 0.5 ug per ml.

Does MOI affect antibiotic resistance?

A: Higher MOI will provide more copies of the antibiotic resistance gene per cell. Cells containing multiple copies of the resistance gene can withstand higher antibiotic concentrations than those at lower MOIs. The concentration of antibiotic should be adjusted to a level that will select for the population of transduced cells you wish to select for, without going below the minimum antibiotic concentration you have established in your killing curve.
