Summary Creative Biogene ensures high-quality lentivirus particles by optimizing and standardizing production protocols and performing stringent quality control (QC). The specific QC experiments performed vary between lentivirus particle lots.
Mycoplasma Creative Biogene routinely tests for mycoplasma contamination using a mycoplasma detection kit. Cell lines are maintained for approximately 20 passages before being discarded and replaced with a new vial of early passage cells. Approximately 2 weeks after thawing, cell culture supernatants are tested for mycoplasma contamination. Creative Biogene ensures that lentiviral products are free of mycoplasma contamination.
Purity Creative Biogene evaluates the level of impurities, such as residual host cell DNA or proteins, in prepared lentiviral vectors to ensure they meet quality standards.
Sterility The lentiviral samples were inoculated into cell culture medium for about 5 days and the growth of bacteria and fungi was tested. Creative Biogene ensures that the lentiviral products are free of microbial contamination.
Transducibility Upon requirement, Creative Biogene can perform in vitro or in vivo transduction assays to evaluate the ability of lentivirus to deliver genetic material into target cells, and assess gene expression and functional activities.
Proviral Identity Confirmation All Creative Biogene lentiviral vectors are confirmed to have correctly integrated provirus using PCR. This test involves transducing cells with serial dilutions of the lentiviral vector, harvesting the cells a few days later, and isolating genomic DNA. This DNA is then used as a template to amplify a portion of the expected lentiviral insert.

The scFv(Control)-CD3zeta CAR-T Lentivirus is a specialized tool designed for the development and validation of Chimeric Antigen Receptor (CAR) T-cell therapies. This lentiviral vector encodes a control CAR construct consisting of a non-targeting single-chain variable fragment (scFv) fused to a CD3ζ (zeta) intracellular signaling domain. Unlike therapeutic CARs that target specific tumor-associated antigens such as CD19 or BCMA, the control scFv in this product is engineered to lack binding affinity for known mammalian proteins. The lentivirus is typically pseudotyped with the VSV-G envelope to ensure broad tropism and efficient transduction of primary human T lymphocytes. By expressing the CD3ζ chain without an active recognition domain, this construct allows researchers to isolate the effects of the intracellular signaling machinery from those triggered by specific antigen-scFv interactions.

This product serves as an indispensable negative control in both in vitro functional assays and in vivo preclinical studies. It is primarily used to establish a baseline for non-specific T-cell activation, cytokine secretion, and tonic signaling inherent to the CAR scaffold. Researchers utilize this lentivirus to differentiate between target-mediated cytotoxicity and potential “leaky” signaling or insertional mutagenesis effects caused by the lentiviral transduction process itself. In multi-color flow cytometry, it functions as a reference for assessing CAR surface expression and persistence. Furthermore, in animal models, the scFv(Control)-CD3zeta CAR-T cells provide a critical benchmark for evaluating the therapeutic window and safety profile of candidate CAR-T constructs, ensuring that observed anti-tumor responses are strictly antigen-dependent. Its use is essential for rigorous data interpretation and for meeting the stringent regulatory requirements for characterizing novel adoptive cell therapies.
