The Tnfrsf9 gene encodes the co-stimulatory receptor 4-1BB (CD137), which is primarily expressed on activated T cells and NK cells. Upon binding to its ligand, Tnfrsf9 activates the NF-κB and MAPK signaling pathways, thereby promoting T cell survival and IFN-γ production. Clinically, Tnfrsf9 is an important target for cancer immunotherapy; 4-1BB agonists can enhance anti-tumor immune responses and prevent T cell exhaustion. Furthermore, the 4-1BB signaling domain is a core component of many second-generation CAR-T cell therapies, providing co-stimulatory signals to ensure the durability and efficacy of CAR-T cells in solid tumors.
