The SQSTM1 gene, located on chromosome 5q35.3, encodes the isolator-1 protein (p62), a multifunctional scaffold protein that plays a central role in the autophagy-lysosome pathway. As a selective autophagy receptor, it attaches polyubiquitinated substrates to the autophagosome membrane. Besides maintaining protein homeostasis, p62 is also a crucial signaling hub in the NF-κB and Nrf2 pathways, regulating oxidative stress and inflammatory responses. Mutations in the SQSTM1 gene are associated with neurodegenerative diseases such as Paget''s disease and amyotrophic lateral sclerosis (ALS). In cancer, p62 accumulation promotes pro-survival signaling.
