Description Monophosphoryl lipid A (MPL), is a chemically modified derivative of lipopolysaccharide that displays greatly reduced toxicity while maintaining most of the immunostimulatory activity of lipopolysaccharide. MPL has been shown to be capable of binding and activating Toll-like receptor 4 (TLR4), present on key antigen-presenting cells, which play an important role in the induction of the innate and subsequent adaptive immune responses. It has been used extensively in clinical trials as a component in prophylactic and therapeutic vaccines targeting infectious disease, cancer and allergies.
